A microRNA-21-responsive doxorubicin-releasing sticky-flare for synergistic anticancer with silencing of microRNA and chemotherapy

A sticky-flare gold nanoparticle probe (AuNP-probe) is designed by the combination of locked nucleic acid functionalized silencing microRNA technology for intracellular microRNA-21 (miRNA-21) sensitively detecting, fluorescence imaging, localizing and silencing. The limit detection as low 0.01 nM. Overexpressed miRNA-21 in cancer cells serves endogenous drug release stimuli to trigger probe-loaded doxorubicin (Dox), which soon translocates into cell nuclei. This multifunctional Dox-loaded AuNP-probe (Dox-AuNP-probe) could induce apoptosis effectively through synergistic effect gene chemotherapy. Dox-AuNP-probe exhibits superior potency compared free Dox molecules, with a inhibition rate 57% (but only 20% Dox) wild-type 30% 0% drug-resistent after 72 h, this strategy not has function sensing, but also can bypass resistance. In MCF-7 xenograft tumor-bearing mice, Dox-AuNP-probes show greater tumor tissues than targeted AuNP-probes (Targeting-AuNP-probe) or molecules. Therefore, represents promising nanotheranostic platform future applications molecular imaging therapy, especially providing potential treat resistant cancers.
